Unit-V: Nonlinear Pharmacokinetics Notes Summary
This unit introduces the concept of nonlinear pharmacokinetics, where drug kinetics do not follow first-order behavior.
1. Introduction:
Nonlinear pharmacokinetics occurs when drug absorption, distribution, metabolism, or excretion processes become saturated.
2. Causes of Non-Linearity:
Includes enzyme saturation, protein binding limitations, active transport, and changes in hepatic or renal function.
3. Michaelis-Menten Method:
A mathematical model used to estimate nonlinear parameters (Vmax and Km), describing how drug concentration affects rate of metabolism.
4. Examples:
Drugs like phenytoin and ethanol exhibit nonlinear kinetics, requiring careful dose adjustment to avoid toxicity
