Anisindione is an oral anticoagulant that prevents blood clots by inhibiting vitamin K–dependent clotting factor synthesis.
Structure of Anisindione
- It is a synthetic derivative of indione with aromatic substitutions enhancing its anticoagulant properties.
- Chemical Formula: C₁₅H₁₅NO₄S
Mode of Action
- Vitamin K Epoxide Reductase Inhibition: Similar to warfarin, anisindione inhibits the enzyme responsible for regenerating reduced vitamin K.
- Coagulation Factor Inhibition: Reduces synthesis of vitamin K-dependent clotting factors II, VII, IX, and X.
- Anticoagulant Effect: Prolongs clotting times, preventing thrombosis and embolism.
Uses
- Thromboembolic Disorders: Prevents and treats deep vein thrombosis (DVT), pulmonary embolism (PE), and stroke.
- Atrial Fibrillation: Reduces stroke risk by preventing clot formation.
- Mechanical Heart Valves: Prevents thrombus development on artificial valves.
- Myocardial Infarction: Lowers the risk of recurrent heart attacks by inhibiting clot formation.
